LAMPIRAN 1
Perhitungan Dosis Ekstrak Air Daun Stroberi (EADS)
Prosedur pembuatan ekstrak air daun stroberi dilakukan di Sekolah Ilmu &
Teknologi Hayati ITB:
1. 500 gram daun stroberi kering ditumbuk menggunakan mortir dan stemper
hingga halus.
2. 300 gram simplisia dimaserasi dengan air suling sebanyak tiga kali (3 liter,
2 liter, dan 2 liter) selama 24 jam.
3. Filtrat disaring dan dipekatkan dengan evaporator pada suhu 70⁰C sampai
ekstrak mengental.
Rendemen ekstrak:
Berat botol
= 83, 3158 gram
Berat botol dan ekstrak
= 177, 6373 gram
Berat ekstrak
= 94,3215 gram
Dosis EADS tikus: 0,06 mg/100 cc, 0,6 mg/100 cc, 6 mg/100 cc, 60 mg/100 cc
(Mudnic, et al., 2009)
Diambil dosis 60 mg/ 100 cc
Rata-rata berat tikus = 200 gram
Volume lambung tikus = 2 cc
Dosis yang digunakan pada tikus:
Faktor konversi tikus 200 gram menjadi mencit 20 gram = 0,14 (Paget &
Barnes,1964)
Dosis yang digunakan pada mencit (20 gram):

Dosis dalam mg/kgBB dihitung dari EADS 3:
EADS 3 =
Jadi dosis yang digunakan:
EADS 1 = 2,1 mg/kgBB
EADS 2 = 4,2 mg/kgBB
EADS 3 = 8,4 mg/kgBB
Cara pembuatan dimulai dari EADS 3:
EADS 1 = 0,042 mg/0,5cc
EADS 2 = 0,084 mg/0,5cc
EADS 3 = 0,168 mg/0,5 cc
Karena kadar dosis terlalu kecil dan keterbatasan dalam penimbangan, maka
diambil dosis tikus 134mg/100cc (Mudnic, et. al., 2009):
Faktor konversi tikus 200 gram menjadi mencit 20 gram = 0,14 (Paget &
Barnes,1964)
Dosis yang digunakan pada mencit (20 gram):
Dosis dalam mg/kgBB dihitung dari EADS 3:
EADS 3 =
Jadi dosis yang digunakan:
EADS 1 = 4,69 mg/kgBB
EADS 2 = 9,38 mg/kgBB
EADS 3 = 18,75 mg/kgBB

Kandungan sildenafil sitrat (1 tablet)
= 50 mg
Dosis efektif sildenafil sitrat untuk mencit = 5 mg/kgBB mencit (Tajuddin, 2005)
Berat badan mencit yang digunakan = 20 gram = 0,020 kg
Volume lambung mencit
= 0,5 cc
Perhitungan dosis yang digunakan
= 5mg
0,020
= 0,1 mg untuk mencit 20 gram
= 0,1 mg/0,5 cc
Perhitungan dosis CMC 1% yang digunakan:
